Notebook covers experiments including:

  1. All Influenza-related experiments initially confirming the sequence (by cloning, experiment 508) of the 54 transcript (see notebook 13).
  2. Experiments (with Ervin Fodor) showing size of poly(A) [32P] ATP-labelled "streak" of a transcript of T7 synthesized 49-mer vRNA (experiment 527) showing polyA was 50 >245 long, with a mid-point at 135 nucleotides long, but may be shorter around 75 nts.
  3. Started experiments (experiment 539) to detect primer-independent synthesized transcripts (previously only detected with ApG priming) of T7-synthesized 49 mer vRNA (from Jane Sharps), initially unsuccessful with γ 32P labelled ATP.
  4. Large scale primer-independent synthesis, with work up, followed by gel analysis with α 32P ATP (experiments 554, 555). First success with primer-independent synthesis in vitro with MN-cores.
