[George Brownlee's son, Edward George Brownlee, died in December 1995 of a brain haemorrhage. See the draft of the obituary for The Times and the Hull Daily Mail at the end of this Notebook and various other notes relating to his death.]

A series of experiments setting up an RT-PCR assay for detecting a full length cRNA transcript of a synthetic 50-mer (tied vRNA panhandle) and MN-cores. Also RT-PCR for polyadenylated transcripts. Both assays were ultimately successful, although unpublished (see below). They provided the basis for further work by David Pritlove and Leo Poon which was published later.

  1. See experiment 453 for sequence of 50-mer.
  2. See experiment 464C for successful poly(A) product detection, a 54-mer using a phased T15A primer.
  3. See experiment 466 for polyA "streak" of an unphased GC clamped T20 primer.
  4. See experiment 485 comparing a T15 and T20 phased primer changing the PCR product from 54 to 59 long. See p. 126 where they are run side-by-side.
  5. See experiment 490 for RT-PCR assay for full length cRNA synthesis, successful at last.
  6. See experiment 492 RNase H targeting of 54-mer.
  7. See experiment 501 PolyA + and – purification of transcripts. Then RT-PCR of PolyA + mRNA and polyA - RNA (i.e. cRNA) separately.
