Dunn School: Notebook 5, May 1985-Jul 1986
File
MS. 12364/6
Held in our offsite storage facility
Notebook covers experiments including:
- With the exception of some unsuccessful cosmid cloning of a human X hybrid (PIP) from Yvonne Boyd (Genetics), most experiments were concerned with the cosmid library of E.coli 803 from which Vroni Knott (new research assistant replacing Joyce Huddleston) and Brownlee picked random clones for Hinf 1 profiling.
- Experiment 392: shows current status of 48 contigs by Hinf1 profiling mainly done by Vroni Knott.
- Experiment 356: shows screening of various E. coli strains from Noreen Murray (Edinburgh).
- Experiment 395: Statistical analysis, theoretically calculating the number of randomly picked cosmids Brownlee would need to analyse the entire E. coli genome.
- Experiments 407, 438: Synthesize gene-specific oligo probes and test cosmids by Grunstein-Hogness hybridization for oligos specific for e.g. DNA Q, Eco GPT, ARG F, LACI etc.
- Experiment 444: Repetitive sequences in E. coli.
- Experiment 445: Current status of 48 contigs (number 47 is missing) covering an estimated 74% of E.coli genome.
- Experiment 451: pDVCosA2 - a new cosmid vector for blunt-ended cloning of sheared E.coli DNA (number 454 library).
- Experiment 457: Joining contigs 5 and 19.
