Notebook covers experiments including:

  1. Many experiments attempting primer-independent synthesis with recombinant flu polymerase using short oligo 5' and 3' promoters. (Experiment 759 onwards). Most experiments were inconclusive, although many experiments suggested there was a different profile of bands when with primer-independent transcription than when a primer, ApG, was used.
  2. Recombinant polymerase prepared using tagged recombinant proteins either PA-His-tagged or PB2 TAP-tagged (experiment 755).
  3. Description of design and construction of control Active Site mutated (ASM) flu polymerase constructed by Tannis Jung (experiment 756).
